I am a novice at this type of research and could use some direction. I am looking to examine any information which indicates the increase in traffic created by the construction of a hotel on Long Island, New York on a site where there is presently a vacant building on a heavily trafficked intersection. I admit I do not know where to begin and could use some direction. Can anyone offer me some, please?

Try Institute of Transportation Engineers (ITE) trip generation rates. You will need to know some basic parameters regarding the "new" hotel, but given that you should be able to come up with the additional traffic that will be added to the street.

Vic- You might also want to give the NYC DOT a call. Try this number: 718 786 4475 They will have recent traffic counts that have been done in the area and you might further commission another count to be done there. The DOT might assist you in this or direct you to have it done yourself.
